Preparation, Purification, and Properties of E. Coli Virus T2
1. A method for the preparation of 8 to 10 liter quantities of T(2) virus lysates, titering 2 to 5 x 10(11) infectious units per ml. has been described. 2. Procedures have been developed for the concentration and purification of virus to a high specific infectivity. No fractionation procedure of the several used succeeded in further raising the specific infectivity of these purified preparation...
متن کاملPurification of TEM-1 beta-lactamase by immunoaffinity chromatography.
A monoclonal antibody prepared against TEM-1 beta-lactamase was found to compete with penicillins and cephalosporins for binding to the enzyme. The purified antibody preparation was linked to Sepharose 4B and used for immunoaffinity-chromatography purification of TEM-1 beta-lactamase. Elution with either benzylpenicillin or cloxacillin yielded a highly purified, concentrated and active enzyme p...
متن کاملSynthesis and Production of Sweet-Tasting Protein in E. coli and Purification by Amylose Resin
A sweet water-soluble protein that reminds stable over wide ranges of temperature and pH, Brazzein has various applications. Its tastes like cane sugar but have no calories. However, the extraction of brazzein from its natural source is expensive and not applicable. In this study we used recombinant DNA technology to provide an alternative option for cheaper mass production of brazzein. A brazz...
متن کاملpH-dependent quenching of the fluorescence of tryptophan residues in class A beta-lactamase from E. coli (TEM-1).
We performed an investigation of the pH-dependent quenching of the fluorescence of tryptophan residues of TEM-1 beta-lactamase from E. coli by uncharged and charged quenchers. pH-dependent Stern-Volmer constants (Ksv/pH) of tryptophan residues allowed us to determine subtle but discrete structurally and functionally important processes.
متن کاملCloning, Expression and Purification of Creatininase From Pseudomonas Pseudoalkaligene KF707 in E. coli.
Creatinine amidohydrolase(EC 3.5.2.10) catalyzes the reversible conversion of creatinine to creatine. Creatininase in combination with other enzymes is used for detection of creatinine in serum and urine which is of significant value for detection of renal, muscular and thyroid functions. The aim of this study was to produce recombinant creatininase enzyme in E.coli expression system to use it ...
